The neuroblastoma is the most common solid pediatric tumor outside the central nervous system. The amplification and expression of the oncogene MYCN in neuroblastoma is associated with a very poor prognosis. The regulation of the MYCN expression is controlled by E2F transcription factors, which can in turn be activated by myc proteins. In addition to previously known E2F-1 activated genes it could be shown that the BMI-1 gene is also regulated by E2F-1. The Bmi1 protein is a member of the polycomb group of proteins that are known to have oncogenic functions. In a second project MYCN promotor-specific proteins could be enriched by means of affinity chromatography. This provides the basis for future characterisation of possible MYCN activating transcription factors.
