According to WHO guide lines seminal leucocytes are quantified in every semen analysis as they can be an indicator of genital tract inflammation. Ten monoclonal antibodies which serve to determine the differentiation and grade of activation of the leucocytes are tested on their qualification to be used in semen. The flowcytometric results of white blood cells in semen are compared to those of peripheral leucocytes. Only the pan leucocyte antibodies CD53 and CD45 as well as the marker for monocytes (CD14) are fit for differentiating white blood cells in semen. Staining of granulocytes is complicated by crossreaction of the antibodies with other semen particles. Seminal lymphocytes can’t be further differentiated into B- and T- cells because of low number. Differentiatin of leucocytes by flow cytometry is much more difficult in semen than in blood. This study shows that not all monoclonal antibodies against white blood cells are able to be used in semen.
