In this experiment 30 trabecular human bone cores were cultivated over a period of 29 days and loaded during 26 days. The mechanical load-signal corresponded to that of physiological jumping and was applied in a unfiltrated as well as filtrated form onto the bone samples. The bone cores were randomly devided into five groups with six samples each and a further subdivision into three male and three female samples. Two groups were terminated using potassium chloride solution. A control-group was contrasted to four load-groups with the following subdivisions: live and unfiltrated signal, live and filtrated signal, dead and unfiltrated signal and dead and filtrated signal. The load signal was applied onto the bone samples with 300 cycles, with a frequency of 1 Hz and an intensity of 3000 μstrain daily. The stiffness (e-module) of each bone core was determined after the actual exposure to the mechanical load while the control-group was only measured every 48 hours. In addition the cultivating medium was exchanged and analysed to bone metabolism as well as to bone cell-specific markers on a 48hour basis. Vitality as well as the activity of the live bone samples were documented via the metabolismic markers, alkaline phosphatase,lactate, lactatedehydrogenase and ph-value, during the entire experiment period.
