Overweight and obesity are increasing challenges for modern society threatening individual health status as well as economical prosperity. Besides conventional treatment a great market for dietary supplements, which promise fast and convenient weight loss, has been established. These pledges have in most cases not been proven scientifically, so in case of the medical devices Matricur™ and CM3™ provided by Dr. Suwelack Skin & Health Care AG and A + G Lifescience GmbH, available since 1999 over-the-counter on the German market. Here we present a first pilot study on these appetite suppressants to reveal possible effects on saturation preception and to uncover a potential way of regulation. We examined 20 healthy probands on three different days, the first day was regarded as control. The other two days three saturation supplements were applied before the test meal. Subjective perception of saturation by using a visual analog scale and gastric emptying by 13C-octanoic acid breath test was measured. Therefore half-time of gastric emptying, lag-periode and gastric emptying coefficient were determined. A hypothetical gut-brain interaction was tested by measuring glucagon-like peptide 1 (GLP-1) and cholecystokinin (CCK) in serum. Our data suggest a positive effect on saturation perception by use of the supplements 30 minutes before food intake. The effect can be seen between 45 and 210 minutes after the test meal. Saturation perception before and during the test meal, as the providers promise, could not be shown. We could also show a significant reduction of gastric emptying time. The half time of gastric emptying was significantly increased compared to the control using Matricur™ by 24% and using CM3-Alginat™ by 26%, respectively. Gastric emptying coefficient and lag-periode showed no siginificant changes. No alteration can also be seen for GLP-1 and CCK. The way of regulation consists of a combination of a mechanical gastric mucosal distension and the nature of the saturation supplements as dietary fibers. We could provide no evidence for a gut-brain interaction. Whether Matricur™ and CM3-Alginat™ contribute positively to obesity treatment can not be verified. Further application studies are needed to proove if saturation supplements help to reduce food intake and achieve weight loss.
