Table of Contents: Type II transmembrane serine proteases (TTSP) are a new and expanding family of proteolytic enzymes. Currently little is known about their physiological functions, although it was shown that many TTSP are overexpressed in human tumors. In this study the expression and regulation of four TTSP (namely MT-SP1, Hepsin, TMPRSS-2, TMPRSS-4) on human intestinal epithelial cells using a real-time PCR was rewied. It could be shown that TTSP and PAR-2 are coexpressed in human gastrointestinal tissues and this with highest levels around coecum. In further inflammatory cell model studies using cytokines and different bacterial strains TTSP expression was detected. Interestingly Entercoccus faecium induced a considerable downregulation of expression. In contrast E. coli Nissle as a probiotic strain induced a strong overexpression. Different preparations of E. coli exhibit that the heat-inactivated preparation induced highest overexpression.
