The aim of this thesis was the establishment of the phage-display technique in the lab of Dr. J. ter Meulen. An antibody-phage-display library was generated from the lymphocytes of a lassa and plasmodium falciparum seropositiv donor. From this monoclonal antibodies in the form of scFv displaying phages should be isolated targeting model antigens (lassa virus, lassa virus GP2, plasmodium falciparum infected erythrocytes and plasmodium falciparum Pff2). The generated library consisted of 5,25 million clones and was thus quite small. Enrichment of antibodies against infected erythrocytes only could beshown, however the original library showed bindung to lassa virus and Pff2 in ELISA and to infected erythrocytes in flow cytometry.
