Aim of the work was the development and validation of a routine method for the detection of the plasticizers Di(ethylhexyl)phthalate (DEHP) together with its primary and secondary metabolites Mono(ethylhexyl)phthalate, ethylhexanol, phthalic acid and ethylhexanoic acid. The analysis was performed by gas-chromatography mass spectrometry after fluid-fluid extraction and derivatisation with MBDSTFA. After evaluation, the method was tested in a small pilot project comparing the exposure of plasticizers in patients undergoing haemodialysis and a control group. Almost all analysts were detected to be higher in haemodialysis patients. The only exception was Mono(ethylhexyl)phthalate, with a slightly lower amount Besides the analytical part, experiments on the immunotoxicological effect of DEHP were carried out on in vitro basis with exposure experiments on Jurkat E6.1 lymphocytes, as well as in vivo experiments investigating the effect in the lungs of DEHP exposed BALB/c mice after short-term and long-term exposure.
