Flavivirin or the NS2B-NS3 protease is an enzyme essential for the propargation of all Flavivirus species. In this work, the possibilities of inhibiting the Flavivirin of the Zika virus (ZIKV) and the West Nile virus (WNV) with cyclic peptide-derivatives were investigated. For this purpose, more than 100 compounds were synthesised and characterised in enzyme kinetic assays, X-ray crystallography and cell culture experiments. The observed structure activtiy relationships may serve as new starting points of the development of antiviral drugs.
