The in-vitro effects of Carboplatin and Lonafarnib on the human Ovarian Cancer cell lines BG1 and SKOV-3, were investigated by proliferation assays, cell cycle distribution analysis, apoptosis induction assay and drug accumulation assys. The proliferation of both cell lines were inhibited in a time and dose dependent manner, calculation of the interaction index revealed drug synergism at distinct drug concentration combinations. Cell cycle distribution analysis showed an arrest in G2-M-phase of SKOV-3. Apoptosis induction assays showed an increase of apoptosis and necrosis in treated cells. Drug accumulation with Daunorubicin showed an increased accumulation after treatment with Lonafarnib alone and in combination with Carboplatin. Carboplatin alone lead to a decreased drug accumulation in the used cell lines. In conclusion it can be said that there are synergistic effects of Lonafarnib and Carboplatin in Ovarian Cancer cells in-vitro.
