Der Replikationsursprung des zweiten Chromosoms, ori2, bildet eine ideale Grundlage für die Entwicklung eines synthetischen sekundären Chromosoms in monochromosomalen Bakterien wie E. coli. Die Regulation der Initiation der DNA-Replikation, vor allem des zweiten Chromosoms, und die Koordination der Replikation beider Chromosomen in V. cholerae sind jedoch noch immer nicht vollständig verstanden. Diese Arbeit soll einen Beitrag zur Erweiterung dieses Verständnisses leisten. Die Koordination der Replikation von Chr1 und Chr2 in V. cholerae erfolgt durch die chr2 replication triggering site crtS auf Chr1, da Replikation der crtS die Initiation der DNA-Replikation an ori2 induziert (Val et al. 2016). Die Kopienanzahl ori2-basierter Replikons, wie des synthetischen sekundären Chromosoms synVicII, ist in E. coli nach Integration der crtS in das Hauptchromosom erhöht. Dieser Effekt konnte genutzt werden, um die Funktionsfähigkeit der crtS verschiedener V. cholerae-Stämme zu untersuchen. Außerdem konnte gezeigt werden, dass die crtS in den Vibrionaceae konserviert ist. Die crtS und ori2 verschiedener Vibrionaceae sind in E. coli bis auf wenige Ausnahmen kompatibel. Zusätzlich zeigten alle untersuchten Vibrionaceae sowohl den aus V. cholerae bekannten Replikationsverlauf ori1-crtS-ori2 als auch eine zeitgleiche Termination der Replikation beider Chromosomen. Weiterhin konnte eine Methode zur Synchronisation der DNA-Replikation in V. cholerae durch Serinhydroxamat (SHX) etabliert werden. Im Zuge dessen wurde der verwendete Stamm V. cholerae A1552 vollständig sequenziert und annotiert. Behandlung von V. cholerae mit SHX führt zur Induktion der Stringent Response (SR) (Haralalka et al. 2003). Im Gegensatz zu E. coli zeigt V. cholerae während der SR einen genau definierten Ablauf von Zellteilungen und Re-Initiationen der DNA-Replikation. Dieser Ablauf wird durch die SR-induzierte Proteinbiosynthese und damit verbundenem Zellwachstum gesteuert und ist somit abhängig von der Produktion des Alarmons (p)ppGpp und dessen Regulation der Transkription. The regulation of initiation of DNA replication, especially of the secondary chromosome, and the coordination of replication of both chromosomes in V. cholerae is still not fully understood. This work should contribute to this understanding. The coordination of replication of Chr1 and Chr2 in V. cholerae is maintained by the chr2 replication triggering site crtS, as replication of the crtS induces initiation of DNA replication at ori2 (Val et al. 2016). The copy number of ori2-based replicons, like the synthetic secondary chromosome synVicII, is increased in E. coli after integration of the crtS into the main chromosome. This effect could be used to analyze the functionality of crtS of different V. cholerae strains. It was shown that the crtS is conserved among the Vibrionaceae. The crtS and ori2 of different Vibrionaceae are, with few exceptions, compatible in E. coli. All analyzed Vibrionaceae showed the order of replication ori1-crtS-ori2, known from V. cholerae, and synchronous termination of replication of both chromosomes. Additionally, a method for synchronization of DNA replication in V. cholerae using serine hydroxamate (SHX) was established. During the course of these experiments, the used strain V. cholerae A1552 was completely sequenced and annotated. Treatment of V. cholerae with SHX leads to induction of the stringent response (SR) (Haralalka et al. 2003). In contrast to E. coli, V. cholerae shows a defined program of cell division and re initiation of DNA replication during SR. This program is controlled by the SR-induced protein biosynthesis and the resulting cell growth and is therefore dependent on the production of the alarmone (p)ppGpp and its regulation of transcription.
