Zusammenfassung:
Die tertiäre Schwefelverbindung Dimethylsulfoniopropionat (DMSP) wird von marinen
Organismen (z. B. Algen) und wenigen Landplanzen in hohen intrazellulären
Konzentrationen synthetisiert und weltweit werden jährlich 109 Tonnen hergestellt. DMSP
dient den Produzenten als Osmolyt und gelangt z. B. durch Lyse der Zellen in Folge einer
viralen Infektion ins Meerwasser. DMSP ist als Vorläufermolekül von Dimethylsulfid (DMS)
ein wichtiger Bestandteil des globalen Schwefelkreislaufes und nimmt indirekt Einfluss auf
das weltweite Klima. Mitglieder der Gattung Bacillus kolonisieren eine Vielzahl
unterschiedlicher Habitate (u. a. marine Sedimente) und haben daher Zugang zu DMSP. Eines
der best untersuchten Mikroorganismen aus der Gattung Bacillus ist B. subtilis. Ein
Forschungsschwerpunkt von B. subtilis liegt auf seiner Anpassungsfähigkeit (z. B.
Osmoregulation) und Adaption an sich stetig verändernde Umweltbedingungen.
Mit einem definierten Set von B. subtilis Stämmen wurden die protektiven Eigenschaften von
DMSP als kompatibles Solute bei Salz-, Kälte - und Hitzestress aufgedeckt. DMSP wird von
B. subtilis nicht verstoffwechselt, sondern dient einzig als kompatibles Solut. Auch natürliche
und synthetische Derivate zeigten Osmoprotektion und teilweise Kälteprotektion bei
gestressten B. subtilis Zellen. Wie bereits für andere kompatible Solute gezeigt wurde nimmt
auch DMSP und seine Derivate Einfluss auf die de novo Synthese des intrazellulären Prolin
Pools und auf das osmotisch induzierte opuA Operon. DMSP und seine Derivate reprimieren
die Expression der Gene für die Prolinbiosynthese und die Expression des OpuA Transporters
bei hoher Osmolarität des Wachstumsmediums. Die Akkumulation von DMSP in allen
Stressbedingungen erfolgte über die ABC Transporter OpuA und OpuC, wobei sich OpuC als
Hauptransporter von DMSP und seinen Derivaten herausstellte. Der OpuC Transporter besitzt
ein breites Spektrum an kompatiblen Soluten die durch das extrazelluläre Subtratbindeprotein
(SBP) OpuCC gebunden werden. Mit Bindungsassays und einem in silico Modell konnte die
Bindung von DMSP charakterisiert werden. Die Subtratspezifität des OpuC Transporters ist
bemerkenswert, während der OpuB Transporter nur Cholin transportiert. Der OpuB
Transporter ist aus einer Genduplikation des OpuC Transporters im Laufe der Evolution
enstanden und hat sich auf die Aufnahme von Cholin spezialisiert. Mit bioinformatischen
Analysen, Mutagenese der extrazellulären SBP von OpuBC und OpuCC und mit
Kristallisationsstudien des mutierten OpuBC Proteins wurde die molekulare Anpassung im
Hinblick auf die Substratspezifität der OpuB und OpuC Systeme beschrieben.
