Zusammenfassung:
Pflanzen produzieren eine Fülle von Metaboliten mit geringer molekularer Masse. Nur wenige gehören zum primären Metabolismus, den alle Organismen teilen, der Rest wird dem Sekundärstoffwechsel zugerechnet. Die Biogenese der Sekundärmetabolite ist von der begrenzten Zahl der Primärmetabolite abzuleiten. Weiter sind Sekundärmetabolite zahlreich und in allen höheren Pflanzen zu finden. Dadurch kann das Vorkommen und die Biosynthese von Sekundärstoffen eine phylogenetische Ordnung von Pflanzen ermöglichen.
In dieser Arbeit wurden zwei Enzyme der Biosynthese von Caffeoylshikimisäure und Rosmarinsäure untersucht. Rosmarinsäure macht den Hauptbestandteil der sogenannten Labiatengerbstoffe aus. Sie besitzt antivirale, antibakterielle und antiinflammatorische Eigenschaften. Caffeoylshikimisäure hingegen ist eine Vorstufe der Monolignole. Monolignole wiederum sind Grundbausteine des Lignins.
Die Ausgangssubstanzen dieser beiden Sekundärmetabolite sind die Aminosäure L-Phenylalanin und, ebenfalls aus dem Skimatweg, L-Tyrosin bzw. Shikimisäure. An der Biosynthese der Rosmarinsäure sind insgesamt acht Enzyme beteiligt, darunter auch die Hydroxycinnamoyl-CoA:Hydroxyphenyllactat Hydroxycinnamoyltransferase oder Rosmarinsäuresynthase (RAS). An die Stelle der RAS tritt bei der Biosynthese der Monolignole die Hydroxycinnamoyl-CoA:Shikimat Hydroxycinnamoyltransferase (HST). Diese Hydroxycinnamoyltransferasen (HCT) gehören zur Proteinsuperfamilie der BAHD-Acyltransferasen, die im Pflanzenreich ubiquitär verbreitet sind. BAHD-Acyltransferasen sind monomere Enzyme mit einem Molekulargewicht von 48-55 kDa. Die Abkürzung BAHD leitet sich aus den ersten biochemisch charakterisierten Enzymen der Familie ab. HCTs katalysieren sehr ähnliche Reaktionen und weisen untereinander eine recht hohe Sequenzähnlichkeit auf. Fast alle bekannten Mitglieder der BAHD-Familie enthalten das an der Katalyse beteiligte HXXXDG-Motiv im mittleren Bereich des Proteins und das in der Nähe des C-Terminus lokalisierte DFGWG-Motiv.
Im Verlauf der vorliegenden Arbeit konnten putative RAS- und HST-Teilsequenzen aus verschiedenen, nahe verwandten Pflanzenarten gewonnen werden. Die putativen Teilsequenzen haben eine ungefähre Länge von ca. 220 Aminosäuren und befinden sich zwischen den stark konservierten Motiven HXXXDG und DFGWG. Putative RAS-Teilsequenzen wurden ausschließlich aus Pflanzen der Familie der Lamiaceae isoliert. Putative HST-Teilsequenzen wurden hauptsächlich in Pflanzen der Familien der Lamiaceae und Boraginaceae gefunden. Die Teilsequenzen wurden verwendet, um phylogenetische Stammbäume, u.a. auch zusammen mit bereits bekannten HCTs zu erstellen. Der berechnete phylogenetische Baum wurde in drei Cluster unterteilt. In den jeweiligen Clustern befinden sich die putativen RAS- und HST-Teilsequenzen. Die Ergebnisse deuten darauf hin, dass beide Synthesewege eine alte Fähigkeit darstellen, die sich in Abhängigkeit von einem gemeinsamen Ursprung entwickelt haben. Für eine konkrete phylogenetische Aussage ist die Anzahl der Taxa jedoch sicherlich unzureichend. Es konnten aber Tendenzen und evolutionäre Unterschiede abgelesen und bewertet werden.
Weiter wurden konservierte Sequenzmotive innerhalb der Teilsequenzen identifiziert. Innerhalb der putativen RAS- und HST-Teilsequenzen konnten dabei mehrere unterschiedliche Sequenzmotive identifiziert werden. In Modelldarstellungen ist ersichtlich, dass diese Sequenzmotive hauptsächlich um den Kanal des aktiven Zentrums angeordnet sind. Die konservierten Sequenzmotive der Teilstücke wurden mit bereits bekannten Motiven abgeglichen und Theorien über deren mögliche biochemische Eigenschaft diskutiert.
Außerdem konnte eine bisher unbekannte Volllängensequenz aus Beinwell (Symphytum officinale, Boraginaceae) isoliert werden. Hierbei handelt es sich wahrscheinlich um eine Hydroxycinnamoyltransferase, die jedoch nicht exprimiert und biochemisch charakterisiert werden konnte. Die Volllängensequenz wurde in einen evolutionären Kontext gesetzt, in silico modelliert und die Daten entsprechend ausgewertet. Daten in Bezug auf Sequenzvergleiche deuteten zuerst auf eine putative HST hin. Im weiteren Verlauf der Arbeit zeigte sich jedoch bei der Berechnung von Phylogrammen unterschiedlicher Sequenzen, dass die Sequenz aus Symphytum officinale eher in die evolutionäre Entwicklung der RAS-Enzyme einzuordnen ist. Die Sequenz scheint sich sehr früh im evolutionären Kontext innerhalb der RAS-Gruppe entwickelt zu haben. Sie ist die erste Sequenz, die sich von den übrigen RAS-Sequenzen an einem eigenen Ast abkoppelt. Sie befindet sich somit auch in direktem verwandtschaftlichem Verhältnis zu der HST-Gruppe und könnte eventuell eine evolutionäre Verbindung zwischen der HST- und der RAS-Gruppe herstellen. Hier könnte eine baldige biochemische Charakterisierung sehr viel Klarheit schaffen und damit die getroffenen Annahmen in dieser Arbeit faktenbasiert analysiert werden.
