Zusammenfassung:
Spannungsgesteuerte Kaliumkanäle (Kv-Kanäle) sind an der Regulation neuronaler und kardialer Aktivität beteiligt. Sie öffnen bei Depolarisation der Zelle und leiten unter physiologischen Bedingungen Kaliumauswärtsströme, wodurch sie zur Repolarisation beitragen. Kv-Kanäle modulieren die Dauer von Aktionspotentialen und die Erregbarkeit der Zellen. Verschiedene Regulationsmechanismen können die Kanalfunktion beeinflussen. Zu den physiologischen Regulationsmechanismen gehören unter anderem die durch endogene mehrfach ungesättigte Lipide induzierte Inaktivierung von Kv-Kanälen, die RNA Editierung des Kv1.1-Kanals, welche zu einem Austausch von Isoleucin gegen Valin innerhalb der zentralen Pore führt sowie die Inaktivierung durch akzessorische Untereinheiten. Sowohl der zugrunde liegende Mechanismus der durch Lipide induzierten Inaktivierung als auch die Folgen der Kv1.1 RNA Editierung sind bisher nur ansatzweise bekannt und wurden in der hier vorliegenden Arbeit genauer untersucht. Zudem wurde die Konformation des Inaktivierungspartikels der akzessorischen Kvβ1.3-Untereinheit und die Regulation der durch sie eingeführten A-Typ Inaktivierung analysiert. Zunächst wurde der Mechanismus der Inaktivierung durch endogene Lipide wie Arachidonsäure, Docosahexaensäure und Anandamid anhand verschiedener Methoden untersucht. Die Charakteristika der Inaktivierung wie Kinetik, Kompetition mit dem Porenblocker TEA und Beeinflussung durch Mutationen in der zentralen Pore sowie die in silico Modelle zeigen, dass eine physikalische Blockade der offenen Kanalpore den wahrscheinlichsten Mechanismus der Inhibition durch Lipide darstellt. Untersuchungen der funktionellen Folgen der Kv1.1 RNA Editierung ergaben, dass editierte Kanäle eine verminderte Sensitivität gegenüber Porenblockern aufweisen. Dazu zählen sowohl die Substanzen 4-Aminopyridin (4-AP) und Psoralen-4, welche experimentell und klinisch genutzt werden, als auch die in dieser Studie neu als Porenblocker identifizierten Lipide und deren verwandte Substanzen. Zudem konnte gezeigt werden, dass editierte Kv1.1-Untereinheiten durch Heteromerisierung die Pharmakologie und Lipidsensitivität aller Kv1-Kanäle modulieren können. Im Rahmen der funktionellen Charakterisierung von editierten Kv1.1-Kanälen wurde in verschiedenen Expressionssystemen eine Verringerung der Stromgröße als Folge der Editierung beobachtet. Daraufhin wurde die molekulare Grundlage dieser Stromverringerung untersucht. Messungen auf Einzelkanal- und Ganzzellebene sowie Quantifizierung der Proteinmenge ergaben, dass die Editierung weder Veränderungen im Öffnungs- und Schließverhalten noch in der Gesamtmenge der Kanalproteine zur Folge hat. Es konnte jedoch eine signifikant verminderte Oberflächenexpression der editierten Kv1.1-Kanaluntereinheiten nachgewiesen werden, die wahrscheinlich für den geringeren Gesamtstrom verantwortlich ist. Diese Ergebnisse beschreiben erstmals eine Modulation des intrazellulären Transportes der Kv1.1-Kanaluntereinheiten durch RNA Editierung. Eine pathophysiologische Relevanz der RNA Editierung des Kv1.1-Kanals wird in Zusammenhang mit dem Krankheitsbild der Epilepsie vermutet. Eine Quantifizierung der Kv1.1 Editierungsrate im entorhinalen Cortex chronisch epileptischer Ratten des Kainat-induzierten Tiermodells zeigte eine erhöhte Editierungsrate im Vergleich zu gesunden Kontrolltieren. Die epileptischen Tiere zeichnen sich durch eine Resistenz gegenüber 100 µM des iktogenen Kv-Kanalblockers 4-AP aus, einer Konzentration, die in gesunden Kontrolltieren epileptiforme Potentiale auslöst. In der vorliegenden Arbeit wurden die Auswirkungen der erhöhten Editierungsrate auf diese Konzentration an 4-AP anhand elektrophysiologischer Versuche im Expressionssystem untersucht. Während eine Beeinflussung des Zeitverlaufes oder der Spannungsabhängigkeit des 4-AP Blocks durch die beobachtete Erhöhung der Editierungsrate ausgeschlossen werden konnte, wurde eine signifikante Verminderung der 4-AP Sensitivität festgestellt. Diese, auf der erhöhten Editierungsrate beruhende, verminderte 4-AP Sensitivität von Kv1-Kanälen liegt vermutlich der veränderten 4-AP Sensitivität der epileptischen Tiere zugrunde. Neben der RNA Editierung wurden im Rahmen der hier vorliegenden Arbeit Aspekte der Inaktivierung von Kv1-Kanälen untersucht. Kvβ1-Untereinheiten können nach Assoziation mit Kv1-Kanälen eine schnelle A-Typ Inaktivierung hervorrufen. Anhand elektrophysiologischer Versuche und Molecular Modelling wurde die Konformation bzw. der Bindungsmodus des Inaktivierungspartikels der Kvβ1.3-Untereinheit analysiert. Die Ergebnisse zeigen, dass der Kvβ1.3-Inaktivierungspartikel während der Bindung innerhalb der zentralen Pore des Kv1.5-Kanals am wahrscheinlichsten in einer Haarnadelkonformation vorliegt und die Inaktivierung durch Phosphoinositide reguliert werden kann.
